Number of Papers published in peer-reviewed journals: Number of Papers published in non peer-reviewed journals: Final Report: High Stability Metal-Protein Interactions Evaluated by Microcalorimetry Report Title Isothermal titration calorimetry (ITC) is a microcalorimetry technique used to study interactions between biomolecules and ligands in aqueous solution. This ARO equipment grant was provided to enable the purchase of an ITC instrument, foster the development of ITC expertise at the recipient institution, and examine the interactions of divalent metal cations with specific proteins using microcalorimetry techniques. An ITC instrument was purchased, specifically the model PEAQ-ITC from the MicroCal division of Malvern Instruments. This is a new ITC model released for sale in 2015, and therefore the PEAQ-ITC had not been tested in the field prior to that date. As an "early adopter" working with a new instrument, the PI conceived and implemented two modifications to the base unit that greatly enhanced instrument reliability: (1) installation of a custom module to improve the vacuum system, and (2) adjustment of a parameter in the instrument control software. After these modifications, the ITC has been operating perfectly. A team consisting of the PI, graduate, and undergraduate students has successfully used the ITC with divalent metal cations and model compounds to obtain stoichiometry and dissociation constant information under various conditions.
